In a world first, scientists are using the controversial CRISPR technology to edit genes while the DNA is still inside a person's body.
Typically, the tool - used to treat conditions like sickle cell disease - removes cells from the body, edits out the mutated parts and then reinserts them, along with instructions to create the proper genes.
But a team at Editas Medicine, a biotechnology company in Cambridge, Massachusetts, says it believes it can use CRISPR like a mini surgeon to slice out non-functioning genes in the body rather than in a lab.
NPR exclusively spoke with two of the four patients undergoing the landmark study, both of whom have been blinded by a rare genetic disorder, with the hopes that the technology will help them regain their sight.

Patients Carlene Knight and Michael Kalberer were both born with a rare genetic eye disorder called Leber congenital amaurosis (LCA).
Children born with the condition have rods and cones - cells that detect dim and bright light, respectively - in the retina that do not function properly.
Some are blind at birth while others experience visual loss over time.
LCA is estimated to affect between one and two babies out of every 100,000 births, according to the National Organization for Rare Disorders.
Both Knight and Kalberer have a version of the disease that is caused by a defect in the gene CEP290, which provides instructions for making a protein present in many cells, including cones and rods.
'I have no peripheral vision whatsoever,' Knight, 54, from Portland, Oregon, told NPR.
'That means I cannot see off the side at all. I can only see straight ahead about the size of a pencil lead.'
Kalberer, 43, from Long Island, New York, says his vision is similar to Knight's, like looking through a tiny window that often blurs.

The tourist was detained at the immigration facility in the airport while police are investigating whether she had committed an illegal surrogacy arrangement. Police Major General Weerapol Charoensiri commander of the Immigration Division 2 department said the campaign against illegal international surrogacy had been stepped up after foreigners, mostly from mainland China, were reported to have taken more than 500 locally born babies out of the country. He said: 'The search for illegal international surrogacy has been intensified. More than 500 locally born babies were taken out of the country by foreigners who were mostly mainland Chinese. 'The cases mostly involved foreign men who married Thai women as proven by marriage certificates. They were trying to take the babies out of the country.' After the reported incidents, the Immigration Division commanded added that they had become more careful when issuing travel documents for infants with suspicious backgrounds. He said: 'We caught the woman through these strict procedures and we are now trying to locate other possible accomplices.' The woman will still be detained at the facility during the investigation. Footage shows the giant balloon slowly descending into a road when it tangled with the cables in Turan district, Shymkent City on May 6. Onlooker Nurzhan Baibakhov said no one was hurt in the incident as the basket was empty and the motorists were able to evade the balloon. He said: 'We noticed something was wrong from distance because it was heading to the road. The drivers stopped before it could crash on them but then it hit the power lines.' A repair team was sent to the area to clear the balloon and began fixing the affected cables and restored the electricity after 24 hours. The local police arrived to investigate the incident and found that the balloon was owned by a businessman who lived nearby. One of the officers told local media. He said: 'The hot air balloon was reported to have been blown away by a strong gust of wind. The owner was not able to control it then before it reached the road.' The charity Bangkok Community Help collects food such as eggs, rice and noodles before delivering it by hand to homes in the Khlong Toei area of Bangkok, Thailand. Health officials say the notorious slum has been a breeding ground in the recent outbreak of coronavirus cases, which has seen daily cases in the country surge above 2,000 in recent weeks. Ministers have started a mass vaccination and testing spree in the poverty-stricken slum - where families cram into tiny wooden shacks - to try and control the outbreak. However, the lockdown has meant that many of the residents who rely on hand-to-mouth jobs for their next meal are going hungry. Wider restrictions on businesses and tourism have seen their incomes wiped out. Volunteers in masks and hazmat suits now risk their own health by walking through the Covid-19 danger zone to take food to struggling locals. Speaking in the video, volunteer Friso Poldevaart, originally from the Netherlands, said: 'We started these organisations when the first Covid-19 wave hit about a year ago. We saw that a lot of people were having immediate trouble, losing their job, losing their income, immediately losing the ability to take care of their families. 'That's why we started to hand out food almost every day, medicine, milk for the kids, dried food, hot food, all things that can help these people through this difficult time. 'Right now we're just trying to help as many people as we can because of this [latest] super severe Covid wave, but our longer-term goals are to teach the kids, build houses for people, help individual families, so hopefully when Covid subsides we will go back to more longer-term things.' The organisation also works with Korranit Ngamsukonratana, a representative who is responsible for the area to provide sirens, speaker, communication equipment, oxygen tanks, blood pressure machines, splints, transport bed, sleeping accommodation, hazmat suits, and other necessary items for the emergency to the locals. Medicine, masks and care packages are also being delivered to people forced to quarantine in their homes after their relatives tested positive for coronavirus. Testing and vaccinations have expanded in the Khlong Toei slums after hundreds of people there were infected. The first case was identified as an employee from an entertainment venue in Thong Lor area which had a cluster that led to a rising number of patients. The Bangkok Metropolitan Administration claims it will soon have vaccinated all 80,000 people in the Khlong Toei, or Klong Toey, area. 'I do not have visual independence. I do not have visual autonomy,' he told NPR.
'The diagnosis could have broken me. And for a while it did.'
Knight and Kalberer have volunteered for the study, along with two other patients, hoping that using CRISPR can help repair their defective genes and restore their vision.
Technically called CRISPR-Cas9, the process typically involves sending new strands of DNA and enzymes into organisms to edit their genes.
The components are often sent into the body on the back of a harmless virus so scientists can control where they go.
Cas9 enzymes cut strands of DNA, effectively turning off a gene, or remove sections of DNA in a lab and are then replaced with the CRISPRs, which are new sections sent in to change the gene and have an effect they have been pre-programmed to produce.

© Provided by Daily Mail But in this study, scientists say they need to use CRISPR to edit genes within the body because the retinal cells are too fragile to be removed, edit and reinserted, NPR reported.
'This is the very first time that anyone's ever actually tried to do gene-editing from inside the body,' Dr Lisa Michaels, chief medical officer of Editas Medicine, told the media organization.
'We're actually delivering the gene-editing apparatus to the part of the body where the disease takes place in order to correct it.'
For the process, three tiny incisions were made in Knight's left eye and Kalberer's right eye to insert billions of benign virus particles carrying the CRISPR technology.
Scientists hope that CRISPR will then cut out the genetic mutation of the cells that affect the rods and cones, which will then spur the body to produce properly working genes, according to NPR.
While it's too soon to say if the technology is working yet, the team hopes it will either prevent further vision loss or even restore vision.
Knight says she hopes that the technology will help her do everyday activities like read a book or see expressions on people's faces.
'To be able to see my granddaughter - especially my granddaughter's face - it would be huge,' she told NPR.
Kalberer says he wants to be able to see computer screens again and watch his nieces and nephews play soccer.
'Maybe be able to - as romantically poetic as this sounds - see a sunset again, see a smile on somebody's face again. It's the little things that I miss,' he told NPR.
